    This genomic region represents an enhancer that was validated by the STARR-seq (self-transcribing active regulatory region sequencing) reporter assay in ground-state (2iL) and metastable (SL) mouse embryonic stem cells, where it was defined as a C1 class enhancer that overlaps an active chromatin signature. This locus also represents Sox2 regulatory region 2 (SRR2), which is located approximately 4 kb downstream of the Sox2 (SRY-box 2) transcription start site. That sequence functions as an enhancer in neural stem cells. POU class family members and Sox2 protein bind together to that sequence to promote activity. Cyclin dependent kinase inhibitors and empty spiracles homeobox 2 negatively regulate that enhancer, resulting in dampened levels of Sox2 expression. [provided by RefSeq, Jul 2023]
