Shipping and storage
Our IVT mRNA products are stored in a 1 mM sodium citrate buffer (pH 6.4) and can be stored at -80°C for up to 12 months. RNA products are shipped on dry ice and freeze-thaw cycles should be avoided.
Gaussia luciferase, a monomeric enzyme containing only 185 amino acids, is very sensitive to be detected even with low expression. As a secreted reporter, its activity can be measured without lysing the cells. This allows cells to be used for downstream applications and real-time or time-course measurements. In addition, Gaussia luciferase is highly stable due to the presence of multiple disulfide bonds, making it suitable for applications at high temperature or with the presence of specific chemicals.

Figure 1. (A) Expression of HiExpress™ Firefly Luciferase mRNA and other luciferase mRNA in HEK293T cells. Cells grown on a 12-well plate were transfected with 0.5 ug of mRNA per well and luciferase activity was measured at 6 h, 24 h, 48 h, and 72 h post transfection. (B) Luciferase activity measured in adult C57BL/6 mice injected intramuscularly with 30 ug of LNP encapsulated mRNA at 6 h, 24 h, and 48 h post injection. FLuc WT indicates wild-type firefly luciferase. FLuc WT (co) indicates codon-optimized wild-type firefly luciferase. FLuc2 indicates Luc2 firefly luciferase.
Figure 1. HEK293T cells were seeded on a 12-well plate and transfected with 1 ug per well of wild-type Gluc, Gluc from competitor T, or HiExpressTM Gluc IVT mRNA. At 6 h, 24 h, 48 h, and 72 h after transfection, 20 ul of the culture medium was taken to quantify the Gaussia luciferase activity. RLU, relative light unit.
