Anti-hCD19 CAR IVT circRNA

circRNA is a novel, covalently closed circular RNA molecule distinct from conventional linear RNA. Due to its circular structure, it is highly resistant to nucleases and therefore more stable in cells. Validated through in vitro expression, VectorBuilder’s anti-hCD19 CAR IVT circRNA encodes a CAR protein consisting of an scFv targeting human CD19, a CD28 transmembrane domain, and a co-stimulatory domain (CD28 or 4-1BB) fused to CD3ζ.

Shipping and storage

Our IVT circRNA products are stored in a 1 mM sodium citrate buffer (pH 6.4) and can be stored at -80°C for up to 12 months. RNA products are shipped on dry ice and freeze-thaw cycles should be avoided.

Experimental Validation

 Expression of Anti-hCD19 CAR circRNA

Figure 1. Western blot analysis of CAR expression in vitro. HEK293 cells were transfected with anti-hCD19 CAR IVT mRNA or circRNA, and cellular proteins were collected 48 hours post-transfection for analysis. GAPDH served as the loading control, and CAR was detected with an anti-CD3 zeta (phospho Y142) antibody. The theoretical molecular weights of h28z and hBBz CARs are 54.26 kDa and 54.15 kDa, respectively. Our results indicate higher CAR expression from circRNA than conventional mRNA.
