    This genomic region represents a DNase I hypersensitive site that is located approximately 20 kb downstream of the beta-globin gene cluster in erythroid cells. It contains an enhancer-blocking element that is mediated by binding of the CTCF transcription factor, and it also includes a silencing element that functions independently of the CTCF-binding site. This locus additionally includes a B cell regulatory element that was identified by FAIRE-seq (formaldehyde-assisted isolation of regulatory elements sequencing), and which was validated as an enhancer by STARR-seq (self-transcribing active regulatory region sequencing) in lipopolysaccharide-activated mouse splenic B cells. Chromosome conformation capture assays indicate that this region is an integral part of the beta-globin active chromatin hub in erythroid cells, where it undergoes looping interactions with the beta-globin locus control region (LCR) and active beta-globin gene promoters.
