HiExpress™ Cre IVT mRNA

CRE recombinase is a ~38 kD Type I topoisomerase encoded by bacteriophage P1 which recognizes two recombinase binding elements (RBE) within LoxP sites to facilitate site-specific recombination. Our HiExpress™ Cre IVT mRNA has been codon optimized to ensure high levels of CRE recombinase expression both in vitro and in vivo.

Experimental Validation

HiExpress Cre mRNA and LNP-mRNA expression

Figure 1. HiExpressTM Cre mRNA and LNP-mRNA expression. (A) Expression levels of WT, Competitor T (m1Psi modified), and HiExpressTM Cre mRNA (m1Psi modified) in HEK293T cells were measured 24 hours post-transfection by Western Blot. (B) Cre-dependent tdTomato expressions in Ai9 mice. Adult male Ai9 mice harboring loxP-flanked STOP upstream of CAG>tdTomato were intravenously injected with either 0.1 mg/kg or 0.4 mg/kg of Cre or HiExpressTM Cre LNP-mRNA to evaluate tdTomato expression. (C) Representative images show liver tdTomato expression 48 hours post-injection.
