HiExpress™ Gaussia Luciferase IVT mRNA

As a monomeric enzyme containing only 185 amino acids, Gaussia luciferase is very sensitive to be detected even with low expression. Unlike other luciferase (e.g. firefly luciferase) expressed intracellularly, Gaussia luciferase is a secreted reporter, which means its activity can be measured without lysing the cells. This allows cells to be used for downstream applications and real-time or time-course measurements. In addition, Gaussia luciferase is highly stable due to the presence of multiple disulfide bonds, making it suitable for applications under conditions of high temperature or the presence of specific chemicals. Its high sensitivity, compatibility and stability make Gaussia luciferase a good reporter candidate for screening various biological processes in vivo and in vitro. Shipping and storage

Our IVT mRNA products are stored in a 1 mM sodium citrate buffer (pH 6.4) and can be stored at -80°C for up to 12 months. The products are shipped on dry ice and should be avoided for freeze-thaw cycles.

Experimental Validation

Gaussia luciferase activity in 293T cells transfected with Gaussia luciferase IVT mRNA and transfection reagent without mRNA.

Figure 1. HEK293T cells were seeded on a 12-well plate at a density of 2.3×10cells per well. Cells were transfected with 1 ug of mRNA per well. At 6 h, 24 h, 48 h, 72 h and 96 h post-transfection, 20 ul of the culture medium was taken to quantify the Gaussia luciferase activity. RLU, relative light unit; Control, cells that were treated with transfection reagent but without mRNA.
