VectorBuilder is the best platform for designing and synthesizing constructs. The online vector design tool is so convenient and fun to use. Ms. Joyce Zhang is always very professional in replying my questions and does it so timely. One time, something did not work out, the refund process was smooth and all the staff were very nice and professional. The constructs I asked for had two fluorescent proteins with two promotors, one has NLS and the other does not. I did transfection and it worked out very well. Joyce helped me all through this process, making my life very easy. Basically, I had many pleasant experiences with Vectorbuilder, and I strongly recommend using it. I had tried other companies, they turned out to be either slow or lacked strong technical support or are expensive. It has been a great experience working with VB. I was one of the first to test their empty capsid (AAV VLP) service. For my batch capsids were nearly completely empty (>97% by AUC and TEM) and were thus a huge help for assay development as a negative control. Our research challenge
Our work involves primary brain cells, including neurons, human astrocytes, and endothelial cells. These cells are notoriously challenging to maintain and to transduce with viruses for gene overexpression or downregulation.

I am very satisfied with the constructions (double mutants) made by Vectorbuilder. The service is very serious and rigorous. They started by verifying our plasmid by doing digestions. As this was not enough, they even sequenced part of the plasmid, which then ensured the success of the constructs. We recently purchased a number of customized plasmids on VectorBuilder. They consisted of expression vectors of two or more ORFs. Our experience was satisfactory, efficient and cost-effective.
The designing stage was facilitated by the VectorBuilder design tool online, in which many of the most commonly used sequences, like fluorescent proteins, selection drug resistance genes, and fusion or cleavage linkers were readily offered to be picked from the ORF lists. Regarding our specific sequences, once pasted into the corresponding window of the design tool, they were immediately matched with the International Nucleotide Sequence Database Collaboration (DDBJ/EMBL/GenBank) accession numbers and compared to the databases. Where there were discrepancies, we were asked which transcripts/cDNAs we preferred to use.
After completing the order, in a matter of three to four weeks, we had in our hands 8 of the 10 plasmids, as offered. For the rest of the
plasmids we provided the template for a specific sequence. Before proceeding with the cloning, they sequenced it, detected a mutation and corrected it at a low extra cost upon our approval. A week later these corrected plasmids were delivered as well. During the whole process, VectorBuilder’s European team kept a personalized email dialogue with us, enabling us to follow-up and make decisions or accept suggestions about the plasmids.We have satisfactorily tested every plasmid now. We think that our decision to work with VectorBuilder has boosted our research by saving us time, resources and enabling us to concentrate more on our experiments.

We utilized two lentiviral vectors, lenti-CYP and lenti-Ctr, from VectorBuilder Inc. in our study on Lgr5+ intestinal stem cells. The lenti-CYP vector, encoding the hCYP27B1 gene, enabled de novo synthesis of locally high 1,25(OH)2D concentrations, proving crucial for our investigation.
