• I would like to share with you my positive experience purchasing a lentiviral vector from VectorBuilder.
   
  First, I am an immunologist and have little experience with cloning. In my recent project, we needed to overexpress a gene in melanoma cells. The transfection efficiency using plasmids in this cell line was extremely low, less than 10%. Then, we tried to use antibiotics to select transfected cells and establish stably expressing cell lines. Unfortunately, for unknown reasons, the ‘stable transfectants’ lost the transfected genes! I was frustrated at this point, until I met VectorBuilder at the EACR conference in Amsterdam. Using the user-friendly website from VectorBuilder, I designed for the first time in my life my own lentiviral vector and got the product in less than one month. My group leader, who has a lot of cloning experience, could not believe that it worked so quickly. Of course, most important are the experimental results. To access the transduction efficiency, I included an EGFP tag in the vector. Now, I can transduce the melanoma cells with nearly 100% positivity. In the following assays, the genes transduced into the tumor cells resulted in a 6-fold induction in tumor immunogenicity.
  Taken together, I strongly recommend VectorBuilder to researchers, especially those who are not so experienced with cloning, and am convinced that the service and product quality from VectorBuilder can further help me to proceed with my own projects more effectively.

• I have been successfully cloning for 3+ years, but the last time we tried generating KO cell lines with CRISPR/Cas9 vectors, we were not successful. This was upsetting, especially since I invested a lot of time in these efforts. Then, by chance, I attended one of VectorBuilder's seminars. Due to their reasonable prices and the fact I could focus my time on other work, we decided to give their lentiviral (dual gRNA) Cas9 plasmid a try.
